The study focused on an enzyme called Caspase-2, which has been a target of interest for treating fatty liver disease. However, the research team discovered that blocking this enzyme might do more harm than good, especially as we age.
According to the lead researcher, Loretta Dorstyn from the Center for Cancer Biology, Caspase-2 plays a crucial role in maintaining the genetic stability of liver cells. It also helps control fat levels in the liver, making it a double-edged sword.
"Liver cells have a unique feature called polyploidy, where they carry extra copies of genetic material. While this can be beneficial for coping with stress, our study shows that without Caspase-2, these extra copies can lead to abnormal growth and damage," Dorstyn explained.
The research team found that mice lacking functional Caspase-2 developed abnormal liver cells with excessive genetic and cellular damage. Over time, these mice experienced chronic liver inflammation, scarring, and oxidative damage, similar to hepatitis-like liver disease. And this is the part most people miss... the aging mice were at a significantly higher risk of developing liver cancer.
"The findings overturn the assumption that inhibiting Caspase-2 is universally beneficial. In fact, this enzyme is essential for removing damaged liver cells as we age. Without it, these cells accumulate and become a fuel source for cancer," Dorstyn added.
